Important cell processes such as cell proliferation and differentiation are regulated by?signalling initiated by fibroblast growth factor receptors (FGFRs). Many evidences showed FGFR signalling involved in serious congenital bone disorders and several types of cancer. However, the biology of FGFR signalling is not fully understood. There are four FGFRs in humans and XX ligands unknown specificity of ligand-receptor interaction in vivo. Although there are studies showing ability of each FGF ligand to activate ERK pathway on overexpressed receptor, these results cannot fully explain the complexity of FGFR signalling. It is not known if FGFRs create homodimers of heterodimers and if cells would respond differently to signalling triggered by one ligand and relayed by FGFR homodimer or heterodimer. Dissecting the precise mechanism of FGF signalling activation is fundamental for development of therapy for treatment of both developmental and cancerous FGFR-related conditions.
