Structural Studies to Elucidate the Phosphorylation-driven Conformational Switch of Dishevelled in Wnt Signaling

Dishevelled (Dvl) is a phospho-protein which plays a central role in Wnt signal transduction. The mechanisms leading to different Wnt signaling pathways through Dvl are still unknown. Dvl has been reported to have open and close conformations due to the interaction between its PDZ domain and C-terminal tail. In vivo data suggests that this conformational switch is controlled by CK1e and NEK2-mediated phosphorylation. Here, we plan to understand the dynamic changes induced by Dvl phosphorylation and the mechanistic details of the conformational switch that define the signal output.
